As KG6BRG noted, try a simplex frequency (no tones, etc.) and make sure the audio input is working correctly on the radio. You can even plug in an FRS or GMRS frequency and test to one of those radios, if you have one.
For your sanity's sake, use the KG-UV Commander
software to do the programming. Using the software, be sure to set the correct RX and TX frequencies, and the "Encode" PL tone (NOT the Decode tone - unless of course your repeater uses one, which is quite rare).
If this doesn't work, you probably just don't have enough power to break the squelch on the repeater. You'll need a better antenna (I've had great luck with a $12 Nagoya NBA-771) or get closer to the repeater.
